Enalees and its technology

Enalees is an innovative company created in December 2015. It is located on the biocluster called Genopole in Evry with 9 employees, the majority of whom are PhDs in molecular biology or biology engineers. A scientific committee composed of a professor in a veterinary school, a PhD in biology specialized in molecular diagnostics and two practicing veterinarians advises about the scientific validity of our projects. Our company is labeled “Hippolia” (the Hippolia Cluster is the only government-certified “competitive cluster” dedicated to the horse industry. It has multiple goals and a single ambition – to position the French horse industry at the forefront of innovation) and is a partner of the French laboratory “LABÉO” which is in charge of the validation of our Epona tests.

The technology used by Enalees is isothermal amplification. This technology allows the amplification and detection of the DNA of the pathogen that is targeted, as when PCR is used, but does not require a thermocycler.

The technology developed by Enalees allows you to confirm the presence or absence of a pathogen in only 30 to 40 minutes after sample collection (10 minutes of sample preparation and 20 to 30 minutes for analysis). The tests can be done directly in your clinic, so you can start a possible treatment of the horse on the day of the test. Epona tests can be used immediately after the beginning of the infection since they directly detect the pathogen’s DNA or RNA.

Technical data

The technology developed by Enalees allows you to confirm the presence or absence of a pathogen in only 30 to 40 minutes after sample collection (10 minutes of sample preparation and 20 to 30 minutes for analysis).

Our Epona tests are validated by our partner LABÉO which, thanks to its expertise in the equine field, was able to evaluate the test performances in comparison with current techniques. The independent technical data generated is accessible to veterinarians.

Equipment and product range

All equipment necessary for the use of Enalees kits (T8 instrument, centrifuge for Epona tests, fixed-volume pipettes) is provided upon installation. In each kit you will find all the necessary consumables for the use of your tests.

For Epona Respiratoire tests you will need large cotton swabs (40cm) that you can buy directly from Enalees (ref 30-0.10-E) or from another supplier.
For Epona Piro-like tests you will need to collect blood with an EDTA tube.

For Astéria Herpès/Calicivirus you will need Enalees cotton swabs (ref 30-1.10-F_EN).
For Astéria Parvovirus blood you will need to collect blood with an EDTA/heparin tube.

The Epona product range is a range of equine veterinary diagnostic aid tests. Enalees offers 18 Epona tests available as panel or individual tests for the detection of pathogens responsible for isolated fever syndrome (Epona Piro-like) and acute respiratory syndrome (Epona Respiratoire) :
– Isolated fever syndrome (Borrelia spp. (Lyme disease), Anaplasma phagocytophilum (anaplasmosis), Leptospira (leptospirosis), Babesia caballi and Theileria equi (piroplasmosis)
– Acute respiratory syndrome (EHV-1 & 4, Equine Influenza and Streptococcus equi subsp. equi (strangles).

The equipment necessary for the use of the Enalees tests (T8 instrument, centrifuge, fixed-volume pipettes) may be made rented or sold by Enalees. By purchasing the T8 instrument, you get lower prices for the purchased tests and no longer pay a “rental fee”.

When clinical signs like hyperthermia are detected in the horse, it may be the case that further to an infection pathogen are present in one or more tissues (blood, pus, urine…). The immune response is not detectable immediately, thus serological tests cannot be used, and only molecular tests, such as Epona tests, are efficient to identify the underlying disease.
Generally, after one to two weeks, and if the animal survives the disease, different types of antibodies begin to appear, making the use of serological tests possible. However, proof of infection can only be obtained by performing two serological tests in a given interval, in order to highlight seroconversion. Therefore, molecular tests are still relevant even at that stage.
Epona tests may be used early in the infection as they detect the pathogen’s DNA or RNA directly.

– Wear appropriate personal protective equipment (gown, disposable gloves changed frequently).
– Once the test has been carried out, discard all the elements of the pouch according to the applicable regulations (pipettes, cotton swabs, preparation tubes and minitubes-reaction). Do not leave the minitubes-reaction in the T8 instrument once the reaction is completed.
– It is recommended to use a flat, clean and clear working surface in a covered and in-door area at a temperature between 15°C and 25°C.
– Do not use a test which pouch has been opened for more than 24 hours, regardless of the storage temperature.
– Do not use a blood tube stored for more than 24 hours and / or stored at temperatures below 4°C or above 25°C.
– Before opening the preparation tubes, make sure that all the liquid or lyophilized pellet contained in the tube is at the bottom of the tube. If necessary, tap the tube several times downwards to bring down any drops.
– Never reopen the minitubes-reaction once inserted into the T8 instrument, or after the end of the reaction. Reopening under these conditions would cause environmental contamination. This contamination is likely to cause false positive results for subsequent tests.

The Astéria product range is a range of feline and canine veterinary diagnostic aid tests. Enalees offers 5 Astéria tests available as panel or individual tests for the detection of pathogens responsible for coryza and enteropathies :

– Coryza (Feline Herpes virus, Feline Calicivirus)
– Enteropathies (Parvovirus)

Samples and manipulations

– Once the test has been carried out, discard all the elements of the pouch according to the applicable regulations (pipettes, cotton swabs, preparation tubes and minitubes-reaction). Do not leave the minitubes-reaction in the T8 instrument once the reaction is completed.

Blood samples must be made in EDTA tubes. The blood must be homogenized before analysis.

The use of an endogenous positive control such as that used in the Epona tests has demonstrated that nasopharyngeal swabs with a large cotton tip (approximately 1 cm in diameter) provide the best quality in terms of sample collection for molecular diagnostic, to the extent that their length reaches the nasopharynx.
Swabs with a thiner cotton tip, designed for genital samples can result in inadequate collection of biological material. This has a negative effect on the overall sensitivity of molecular biology tests (PCR and Enalees), and thus the thin swab should not be used for this type of sampling.
In addition to the type of swabs, the swabbing technique is important. The filthy appearance of the swab is not a reliable indicator of proper sampling. Dust and nasal secretions are easily collected with sometimes only a small part of or not enough biological material. With the nasopharynx just below the horse’s eyes, almost the entirety of the swab should be inside the nasal cavity during swabbing.

Epona kits are kept for up to 1 year after the manufacturing date. They shall be preferably stored in a fridge but can be stored at room temperature without degradation of the quality of the test. The expiry date is indicated on the outside of the Epona box.

Reopening under these conditions would cause environmental contamination. This contamination is likely to cause false positive results for subsequent tests.

It is not necessary to change filter-tips when dispensing DNA into each minitubes.
